This preclinical study evaluated MECS hard capsules, a Vietnamese herbal formula containing Mimosa pudica, Erythrina variegata, Cinnamomum iners, and Scoparia dulcis (40:12:16:32 ratio), for sedative and anxiolytic effects in Swiss albino mice. Researchers employed molecular docking, ADMET prediction, QSAR modeling, and molecular dynamics simulations alongside in vivo testing. UPLC-QToF-MS confirmed 9 of 11 predicted bioactive compounds. Safety assessment showed no adverse effects at maximum dose (39 g/kg) or during 60-day subacute toxicity testing at 346 and 692 mg/kg doses. Behavioral assays using light-dark box and elevated plus maze models demonstrated significant anxiety reduction. Sleep recovery models with diazepam showed prolonged sleep duration and reduced sleep onset latency at both doses. The combined in silico and in vivo evidence supports MECS capsules as a safe herbal intervention for anxiety and insomnia, warranting consideration as adjunctive therapy in integrative clinical practice.
